Learning Objectives

The course aims to train PhD students on an advanced cellular analysis technology: flow cytometry.

In this sense, the course will develop into basic lessons that will allow the doctoral student to understand the rudiments of this technology, in particular the physicochemical bases that allow to study cells and subcellular particles using fluorochromes as specific dyes or adhering to specific vectors (eg. Monoclonal Antibodies) using multiple fluorescence analysis.

Application lessons will follow which will cover the two major fields of application;

a) cell biology (study of cell kinetics, cell stress phenomena, apoptosis, study of cytoplasmic organelles)

b) the study of the phenotype of normal and pathological hematopoietic cells

Program

Physical basis of light (Lasers) and light detection systems (PMT and Photodiodes)

Fluid physics, hydrodynamic structure of the flow cytometer.

Optical bench, pulse generation and their digitization, biparametric dot-plot matrix for the study of signals, gating systems of the clusters highlighted in the matrices and the "Boolean gating" for identifying subsets.

Compensation of fluorescences on the basis of the emission spectrum of fluorochromes

Mechanical and electrical cytometric sorting

Bridging Courses

It is advisable to follow module 1 and module 2 at the same time

Learning Achievements (Dublin Descriptors)

Once the student has followed the course in its 2 modules, he will be able to evaluate the possibility of using this technology and the possible benefits that the application of this investigation procedure will bring to his research project. Furthermore, he will have had the opportunity to see and understand in detail the clinical, immunological and oncological applications of flow cytometry.

Supporting Activities

Upon agreement with the doctoral student, attendance at the cytometry laboratory will be possible

Assessment

A short quiz on the activities carried out will be given at the end of the whole module.
